   Proposal Description:
   The advent of positron emission tomography (PET) ligands that bind to Alzheimer???s disease (AD) hallmark protein amyloid-?? have caused a major paradigm shift in diagnostic thinking. Amyloid imaging has been incorporated in novel diagnostic criteria for various stages of AD (McKhann et al. 2012 Albert et al. 2012 Dubois et al. (2010) Sperling et al. 2012) and is increasingly used as inclusion or exclusion criterion for patients in studies and clinical trials. This is justified by several clinical studies showing that the correspondence between clinical diagnosis and PET findings is modest (Ossenkoppele et al. 2012 Schipke et al. 2012 Grundman et al. 2012). With almost a decade of experience in amyloid imaging, this is the time to review  amyloid imaging studies and establish concordance and disconcordance between  clinical observations and molecular findings on PET with neuropathogical data  from NACC as reference sample.   Our aims are:                                 AIM 1: to estimate the prevalence of amyloid as measured with PET in demented patients.(in a) AD, b) FTD, c) DLB, d) VaD, and e) dementia-other (e.g. CBD, PSP, PDD etc.)                                                                             AIM 2: to relate this prevalence to age, sex, education, APOE genotype and disease severity.
